Broadly Neutralizing Antibodies and Viral Inducers Decrease Rebound from HIV-1 Latent Reservoirs in Humanized Mice

Latent reservoirs of HIV-1-infected cells are refractory to antiretroviral therapies (ART) and remain the major barrier to curing HIV-1. Because latently infected cells are long-lived, immunologically invisible, and may undergo homeostatic proliferation, a "shock and kill" approach has been proposed to eradicate this reservoir by combining ART with inducers of viral transcription. However, all ...

Neutralizing the HIV Reservoir

Halper-Stromberg et al. use a humanized mouse model to demonstrate that broadly neutralizing antibodies, when administered with a combination of HIV latency activators, can reduce persistent HIV reservoirs, as measured by plasma virus rebound. Their results support the use of broadly neutralizing antibodies in HIV-reservoir-purging strategies.

Neutralizing Epitopes of HIV-1

Ideally, a vaccine against human immunodeficiency virus type 1 (HIV-1) would induce neutralizing antibody levels which would provide sterilizing immunity. The levels of antibody required however may not be achievable by vaccination (Gauduin et al., 1997; Parren et al., 1997; Moore and Burton, 1999; Shibata et al., 1999). Nevertheless, neutralizing antibody responses albeit of lesser, but still ...

HIV vaccine design: the neutralizing antibody conundrum.

A brief historical overview of anti-HIV vaccine approaches The identification of the human immunodeficiency virus as the causative agent of AIDS [1–4] was followed by intensive efforts to develop a vaccine against this pathogen. Initial vaccine approaches aimed at the elicitation of anti-viral neutralizing antibodies (NAbs). The sole target of anti-HIV NAbs is the viral envelope glycoprotein (E...

Broadly Neutralizing Antibodies for HIV Eradication.

Passive transfer of antibodies has long been considered a potential treatment modality for infectious diseases, including HIV. Early efforts to use antibodies to suppress HIV replication, however, were largely unsuccessful, as the antibodies that were studied neutralized only a relatively narrow spectrum of viral strains and were not very potent. Recent advances have led to the discovery of a l...
